چکیده

To begin with, Human Pharmacology is a moderately priced book that makes no pretense to be as encyclopedic as Goodman and Gilman. One of the main advantages of the book is its consistency of presentation. Each chapter is presented in five sections: (1) therapeutic overview; (2) mechanisms of action; (3) pharmacokinetics; (4) relation of mechanisms of action to clinical response; and (5) side effects, clinical problems, and toxicity. This arrangement allows the reader to develop a consistent and organized framework for understanding each major class of drugs. Human Phannacology is particularly good in presenting the mechanisms of action of each drug class. The generous use of color and diagrams, combined with concise and simple explanations, adds to the clarity of the presentations. This fact alone may allow the book to stand out, as mechanisms of action are, sadly, presented in a bland and enigmatic manner in most other texts. In addition, the authors make a genuine effort to relate the mechanism of action to the clinical response of the drug, which adds invaluably to the student's appreciation of the subject matter. As I forewarned in my caveat to this review, however, all texts have their shortcomings. Ironically, in Human Phannacology, one of its shortcomings stems from one of its strengths. In trying to present the mechanisms of action in as clear and simple a manner as possible, the authors have risked oversimplifying some of the processes and masked some of the complexities involved that are required for understanding the toxicity of some of the drugs. Despite the general clarity of the other sections in each chapter, the pharmacokinetics section often seems to be the most confusing. It is a hodgepodge collection of details, some ofwhich are important, some of which are not. The section is never divided into absorption, distribution, metabolism, and excretion, even when appropriate, and seldom divided into drug subclasses (e.g., antidepressants into tricyclics, second generation, and monoamine oxidase inhibitors), even if doing so would add greatly to the clarity of the section. In conclusion, I think it is obvious that Human Phannacology should not be used as a reference text. The clear and colorful presentation of its subject matter is obtained at the expense of pharmacokinetic and pharmacodynamic lack of detail. This presentation may, however, appeal to first-time students of the subject. I cannot help but think that this book may not be adequate for medical students who might desire a textbook with greater depth and substance.

Prenatal Diagnosis, Sampling of Chorionic Villi

Prenatal diagnosis with application of the Amniotic fluid (Amniocentesis: AC) and chorionic villi sampling {CVS) is a general aspect of application in Human Genetics which facilitate the diagnosis of different Genetic defects, enclosing chromosomal abnormalities and Genosomopathies. Diagnosis of Genetic defects for AC and CVS is between 16-20th and 8-12th weeks of gestation respectively. Allica...

Fragile X testing in a diagnostic cytogenetics laboratory.

Chromosome results obtained from 1012 patients referred with developmental delay without known cause within the three years 1985 to 1987 are reported. G banding analysis and assessment of 70 cells for fragile X gave abnormal results in 84 cases: fragile X in 31 patients and other abnormalities in 53 patients. A further 16 sibs expressing the fragile X were detected in family studies originating...

the variable expression in families. Of note, however, in the fact that the identification of the defect at the molecular level, recently published by Sherman and others, has made much of the molecular marker information presented here obsolete. Many of the confusing aspects of the reported pedigrees can now be explained by the observation of a variable number of CGG repeats at the fragile X lo...

Translocation t(3;8)(p14.2;q24.1) in renal cell carcinoma affects expression of the common fragile site at 3p14(FRA3B) in lymphocytes.

The common fragile site at 3p14(FRA3B) is cytogenetically close to the positions of translocation and deletion breakpoints frequently observed in renal cell carcinoma (RCC) and small cell carcinoma of the lung. Possible involvement of this fragile site in the familial RCC t(3;8)(p14.2;q24.1) was investigated. Expression of FRA3B, induced by treatment of lymphocytes with aphidicolin, is altered ...

The frequency of chromosomal abnormalities in patients referred for fragile X analysis.

The present paper summarizes our existing database on chromosomal abnormalities found in patients referred because of a question of the Fragile X Syndrome during the period from January 1, 1990 to June 30, 1995. Cytogenetic results were derived from testing performed at the cytogenetics laboratory at Rhode Island Hospital. All positive fragile X individuals detected among our sample population ...

Mechanisms of common fragile site instability.

The study of common fragile sites has its roots in the early cytogenetic investigations of the fragile X syndrome. Long considered an interesting component of chromosome structure, common fragile sites have taken on novel significance as regions of the genome that are particularly sensitive to certain forms of replication stress, which are frequently rearranged in cancer cells. In recent years,...
